Comparative Study of Diuretics & Antimalarial Drugs on QT - Interval Prolongation in Rat

Abstract: Recently, numerous cases of electrocardiographic QT- interval prolongation and subsequently fatal polymorphic ventricular tachycardia termed Torsed de Pointes related to treatment with cardiac and non-cardiac drugs. The drugs are known to belong to different chemical class that may induce long QT syndrome via inhibition of a rapid component of delayed rectifier K+ channels (Ikr) encoded by the human ether-a-go-go related gene encoding mutation often underlies such preclinical and clinical finding. Some drugs like diuretics ( Hydrochlorthiazide and Furosemide) and antimalarial (Mefloquine, Artemether and Lumefantrine) has also been reported to be preclinically and clinically induce long QT Syndrome and further torsade de point and leads to sudden death.
